A Portuguese company operates a bus line from Porto to Madrid; buses are departing from Madrid Avenida America at 08:30am and 10:00pm and from Madrid Estação Sur at 09:30am or 11:00pm to arrive at Porto at 5:30pm or 06:30am. The single regular fare is €48 and the return trip is €84.

But why do you want to take the hardship of a 9 hours bus travel if you could get a 1:20 hour budget flight by ryanair. Planes are departing daily at 5.50am and 8.50pm. If you book early enough you may get a flight for €5.00; the regular fare including taxes and fees is €43.38.

WHY are you flying to Madrid, to get to Oporto???? I’d suggest taking the train, and even still, it’s the better part of a day…at least four or five hours…plus possible transfers…much longer by bus.
How about flying (from Madrid) to A Coruña or Santiago de Compustela? It would get you much closer, but then, the connections might be prohibitive…check that, though. Flights within Spain are quite cheap. Check Spanair.

as someone else has said the fast train to Portugal leaves from somewhere in the North – the Basque trains did a system that allowed you to visit Portugal for a day, for the price of 1 ticket when it first opened last year
try this company ;
